ActivSignal is an American company that transforms technology into systems biology proteomics tools, located in Natick, Massachusetts. The marquee product, the IPAD platform, can determine the activity of multiple signaling pathways in one reaction.

IPAD Platform[edit]

The IPAD platform (Immuno-Paired-Antibody Detection platform) was developed to help academic, pharma and biotech researchers understand the Pathway activation/inhibition profile of cells. This platform assesses the signaling pathway map profile of more than 20 major pathways at once for each sample. The IPAD platform panel includes 70 different human protein targets, measuring protein modifications and expression for each, with coverage of multiple targets per pathway.

The technology uses antibodies against established and well known targets that reflect activities of canonical pathways. The IPAD platform can be viewed as a sophisticated highly multiplexed sandwich ELISA, using proven pairs of antibodies against known phosphorylation sites, e.g. Akt Thr308; cleaved proteins, e.g. cleaved Caspase-3; protein expression, e.g. p21Cip.

The ActivSignal platform is versatile and can accommodate any number of samples from 2 to 96 and above. The assay is performed in a 96-well plate format, thus it is well designed for screening purposes. The IPAD assay can be used to profile samples made from cell culture, frozen tissues, FFPE sections, biopsies including fine needle biopsy, blood, etc. The IPAD assay requires only 150 nanograms, which can be easily taken from samples where available amounts may be limited, e.g. upon laser dissection of tissue.
