Overall, your article is very easy to understand and I think you explain it very well despite having some difficult concepts to discuss. I think some minor things you could consider:

1) Adding a hyperlink to "heterotetrameric" in the 1st paragraph of mechanism of action might be helpful.

2) Adding one sentence about how IGF1R amplification and overexpression leads to excessive cell proliferation? Just to connect the last sentence of the first paragraph to the 2nd paragraph so it's a bit easier to read how IGF1R is involved with tumor growth.

3) Listing out current medications on the market that are also IGF1R targets or like other similar drugs targeting IGF in the History section. Is Dalotuzumab the first drug targeting this receptor?
