the diagram is incorrect - it contains an unnecessary restriction digest post-loop formation. PCR can be carried out on circular DNA!!!

You're right, it's not necessary and should probably be changed. In my experience, however, linearizing circular templates sometimes increases the reaction efficiency, especially with larger products. – ClockworkSoul 17:09, 12 February 2010 (UTC)[reply]

as a scientist who not only does pcr, but was possibly the 1st person at MIT to do pcr, this article is worse then useless, imo - and I know how to talk to non scientists. scrap it and start over The essential point is that one starts with a circular piece of DNA and converts that to a linear with the primers at the end; from that you can talk about uses etc — Preceding unsigned comment added by 50.195.10.169 (talk) 20:52, 18 July 2013 (UTC)[reply]
