EC50 vs IC50

[edit]

I'm puzzled. After creating SR9009, Boghog adds some useful information to the article by quoting the maximum inhibitory concentration from the pubmed article. So I go into SR9011 and add the same information. But as I read the primary source article, I see the contrast of "IC50" and "AGONIST". Wait. This is an agonist. Why are we measuring IC50 and not EC50? This is not an antagonist. Yet, the primary source clearly lists an INHIBITORY CONCENTRATION and not an EFFECTIVE CONCENTRATION. Can someone please provide a clue to me? Foamyslippers (talk) 02:59, 27 August 2013 (UTC)[reply]

I also did a double take when I read the source (PMID 23852339). An agonist is defined as a ligand that mimics the activity of the the endogenous ligand. The endogenous Rev-ErbA ligand is heme which by binding to this receptor down regulates (represses) gene expression. The data that Burris reports are from a Rev-ErbA reporter cell line that measures the repressor activity of Rev-ErbA. Hence this is an "inhibitory" effect for which IC50 is an appropriate designation. Boghog (talk) 09:01, 27 August 2013 (UTC)[reply]
I have edited the article so hopefully it is now clearer. Boghog (talk) 17:43, 27 August 2013 (UTC)[reply]
