In the first section I am confused about the C rich tetramers that form. Possibly elaborating or rewording the sentence that explains what this is could help to clarify. The rest of the structure section is great and provides solid details. In the biological implication section, the beginning is a bit repetitive so possibly condensing or changing some of the phrasing would be good. The application section is great and uses two good and reliable sources. All of the citation links work and the page looks great. Some minor phrase changes and wording will help to make things more clear but overall this page taught me about i-motifs and uses great examples. Well done! Structure Additions

I am a little confused about what an intercalated, anti-parallel parallel-stranded duplex tetramer structure is. It would be helpful if you linked the word to another wiki page that talks about what it is or just mention a little be more about what it looks like in a sentence. Also, I think it would be helpful there was a picture (like from chem draw or pymol) that showed the C-C+ bond with the hydrogen bonds to the amines, oxygens, and nitrogen’s that are talked about. Overall, there was a lot of good structure information and it looks good! I would suggest rereading the paragraph to check for any awkward sentences/writing mistakes. The sentence “It is speculated that the pH-dependency of i-motif the structure is a key part of its in-vivo activity and controls its function" was worded funny.

Biological Implications

In the first sentence I am not sure you need to say, “postulated to potentially….”. Those words together sound redundant. I would just use postulated because it sounds more confident than potentially. I would also list a couple of the proteins/ligands which recognize G/C-rich DNA regions. I am confused by the sentence that says “human DNA was isolated to occur in the late G1 phase of the cell reproduction cycle”? Is it supposed to say “human DNA was found to occur…? I also would like to know a sentence or two more about i-motifs acting as molecular scaffolds.

Applications

I liked the analogy to the trash bag because it helped me visualize what it looked like when the i-motifs form. The Applications section was very straight forward, and I liked how you linked some of the words to other wiki pages that had more information about those words. I am not really sure what else you could add to this section.

Overall, it was really good! There was a lot of good information that you both added, and it was interesting to read.

Additional comments: emphasize that is four-stranded. Hard to picture with your description. what technique was used to show i-motif formation at neutral pH and what system? Why does pH dependency play a role in function? You can condense the information about the requirements and stability for i-motif formation.

Why does the fact that proteins bind to GC rich regions indicated about i-motif?

What is hTelo?

"presence of i-motif structures in human DNA was isolated" Is it possible to isolate i-motif structure? Isolating the presence of? Not clear. increased i-motif expression- what does this mean? normally express a protein.
